An Early Stage Researcher (ESR) position is available in the Horizion 2020 Marie Curie European Training Network (ETN) EUROPOL at the Technical University of Denmark. Nuclear Magnetic Resonance spectroscopy (NMR) and Magnetic Resonance Imaging (MRI) play unique roles in contemporary science, from physics, chemistry and biology, to clinical research and diagnosis. However, further development of both NMR and MRI is hampered by a limitation in sensitivities. This problem is unlikely to be solved by 'bigger machines' given the high levels of development already in this field. This project looks to develop a new approach by combining NMR and MRI with nuclear hyperpolarisation, which produces signals that are 50,000 times stronger than those produced by NMR and MRI. The EUROPOL consortium contains leading experts: in physics and engineering of magnetic resonance, in synthetic chemistry for generating new labelled compounds; in the use of NMR for structural and cell biology; and in the use of preclinical and clinical MRI applications. The project is guided by the concept that only by teaming together these key areas of expertise, can the potential of hyperpolarisation be realised. EUROPOL will provide training to Early Stage Researchers (ESRs) in all the topics underlying the advancement of magnetic resonance research. ESRs' projects will include: advanced physics; development of new instruments, and new ways of exploiting NMR and MRI's hyperpolarisation potential; biophysical NMR; screening of healthy and diseased metabolomes; expanding the available substrates to be targeted by in vivo MR; cell and system biology exploration to investigate the nature of the metabolome; and in vivo hyperpolarisation strategies in MRI. In this particular project Dynamic Nuclear Polarization will be studied as a function of field and temperature for samples relevant to the Europol consortium. Relevant molecules include glutamine, glutamate and glucose. These molecules will be formulated to maximize DNP and enable in vivo experiments. It is important to understand the role of the matrix (deuteration, degassing, different solvents, etc) as well as the effect (efficiency) of the radical. Radical and target molecule concentration effects will be studied, and mechanisms to remove the radical will be investigated. The samples will be studied under a range of field and temperature conditions. DNP will be studied at discrete magnetic fields of 3.35 T (94 GHz), 6.7 T (188 GHz) and 10.1 T (282 GHz) and over a temperature range of 1.2-20 K. The polarization of the various nuclei (e.g. 1H and 13C) within the sample will be studied to explore more efficient polarization schemes such as cross polarization. Comparison between direct polarization with narrow line radicals and cross polarization through protons with broader line radicals will be performed.
