Postdoc in Development of MHC I and II High-Throughput Production Pipelines

A postdoctoral position is available in the ‘T cell recognition and Cancer’ group headed by Associate Professor Sine Reker Hadrup at the Section of Immunology and Vaccinology, at the Technical University of Denmark, Veterinary Institute, Frederiksberg. The ‘T cell recognition and Cancer’ group is currently consisting of 15 people, including Postdocs, PhD students, Technicians and undergraduate students. The overall aim of the research group is to gain novel insight to T cell recognition of cancer and autoimmune diseases, through use and development of novel Technologies. Responsibilities and tasks The postdoc will be responsible for the development of MHC class I and II protein production. A high-throughput platform for MHC class I monomer generation is already running in the lab through UV-induced peptide exchange. We aim to extend this platform to additional MHC class I molecules and also MHC class II. Main focus will be on the design and development of MHC class I and II conditional ligands and a flexible and productive platform for protein expression and purification. MHC class I and II molecules are produced through expression in E.coli, and protein refolding and purification on size-exclusion HPLC. MHC class I and II molecules are the cornerstone of all T cell analyses in the research group. Development of this production platform is crucial. High-throughput platforms for MHC production are of interest for immune assessments in both infectious diseases, cancer and autoimmune diseases. The project is closely related to other ongoing project in the group, and collaboration and shared responsibility across projects in the research team is expected. Qualifications Candidates should have a PhD degree or equivalent. It is important that the applicant has experience with protein purification, expression, refolding, and purification. This includes E.coli based transformation, expression and purification of inclusion bodies. Additionally experience with use of MHC-multimers for T cell detection, and Flow Cytometry is preferred.
