Postdoc: Interplay between environmental and genetic drivers of pancreatic ductal adenocarcinoma

A three-year postdoc position is available, tentatively from Nov 15th 2018 in an interdisciplinary project enabstractJV_titled “Characterization of the interplay between environmental and genetic drivers of pancreatic ductal adenocarcinoma”. Pancreatic ductal adenocarcinoma (PDAC) has an exceedingly poor prognosis and is predicted to become the leading cause of cancer mortality by 2030. Characteristic hallmarks of the disease are several specific driver mutations and a dense, acidic microenvironment, yet the interplay between these factors in driving disease onset and progression is uncharacterized. In this project, an interdisciplinary team will address this question using state-of-the-art organotypic PDAC models and advanced bioinformatics approaches. The long-term goal is to identify early PDAC biomarkers and novel treatment options.

Research environment
The project is a collaboration between Prof. Albin Sandelin, BRIC and Dept. of Biology, and Prof. Stine F. Pedersen, Dept. of Biology. The candidate will be based in Stine F. Pedersen’s group, and will work closely with a postdoc counterpart in the Sandelin lab and a research assistant/technician in the Pedersen lab. The two groups are located within 5 minutes walking distance in the Northern Campus of the University of Copenhagen. Together, the groups employ several postdocs, PhD students, and undergraduate students and provide a strong, international research environment. The project will additionally be affiliated with the Marie Curie ITN project “pHIoniC”, in which two PhD students will be recruited for the project.

Project and specific qualifications
The candidate will be responsible for the organotypic PDAC models, including introduction of relevant mutations, mono- and co-culture in 3D, and acid stress treatments, and for phenotypic measurements at defined time points. These will be matched with parallel DNA/RNA sequencing analysis, performed together with a bioinformatics postdoc counterpart. Finally, organoids driven to PDAC phenotypes by mutations and/or acid treatment will be orthotopically injected in syngeneic mouse models to monitor growth and metastasis compared to un-treated control organoids.

The ideal candidate has strong expertise in advanced mammalian cell culture systems and CRISPR/Cas9 technology and other molecular biology approaches, and broad background knowledge in cancer research. At minimum, the candidate should have an interest in bioinformatics; actual experience in computational analyses is an advantage. The candidate must have a passion for working in a highly interdisciplinary environment.

The candidate must have a PhD in a life science field, with a documented track record of excellent scientific publications. Excellent written and spoken English is a strong requirement. The candidate must the ability to work autonomously as well as in a team.

While negotiation for salary supplement is possible, the starting salary is currently up to 424,526 DKK/year including annual supplement (+ pension up to DKK 72,594).
